Hi Beck,
I found the source:
Web of Life Student's Manual Part 2 11th edition
9b4 Exercise Mitosis is root tips page 31
"Put the onions in a dark cupboard overnight.

- Fixing the Root Material: As early in the school day as possible (and not later than 9 am), take the onion preparation out of the cupboard.

Cut or break off I cm from the end of each root ........... (If the root does not break easily when twisted with forceps, leave it - it is probably too old to show many dividing cells)"

Cheers,
Glenda

Subject: [Catalist] Mitosis experiment

Hi all,

I am a lecturer at a TAFE, and I am seeking any feedback and assistance with experiments related to mitosis.

We have been trying to do the garlic root tip experiment, but with not much success (even though I had a lab technician previously who seemed to get it right, our new lab techs are not having much luck). The root tips grow well, but when we perform the microscopy, we don't see any mitotic phases in the cells. Not sure if we are leaving the tips to grow too long, or some other issue. I have attached the prac procedure.

Any tips, hints or suggestions from you??

Regards,
Beck
